Are supply teachers ever expected to take assemblies?
Again, this is very unusual. If any such specialist role were likely to feature in a placement the school would make a specific request at the time of booking. Furthermore, we would expect the booking to be made with plenty of time for the supply teacher to prepare. In reality, it is only likely that a supply teacher would be asked to take an assembly if s/he had spent a lot of time at that specific school and were familiar with the staff and pupils – it would be unfair to expect anything else.
